Anthony_A:I've lost that amount of weight in 6 weeks.
Gracie Diet and rolling 5 days a week.
Is it just a manner of speech or is there actually a 'Gracie Diet?'

Lechuza -
the Gracie Diet is real, and its just a common sense diet (some say too low in protein but only if your trying to put on muscle mass). It does have some interesting ideas. Do a google search and you should find info.
SAFTA4Life
Lets assume you're serious.
It really just depends on how much fat you have on you, and how unfit you are.
Cut out all saturated fat, refined sugar, wheat, and alcohol NOW. There is no cheat day, there is no treat for doing well. Cut all of it out now.
Stick to fruit, steamed or raw vegetables, oatmeal and/or museli (for breakfast) and lean beef, chicken, and fresh fish.
Add in some protein shakes if you want.
If you have to lose weight fast, which I don't really recommend, you can try one of the fat burners - just make sure its Ephedrine free.
(on a side note, I can't believe that **** is still legal in the US. I'm looking through a US mag at the moment - the UFC Muscle and Fitness edition, and all the Ephedrine based fat burners now seem to have the warning: may cause serious adverse side effects, not to be taken by persons under 18....for **** sake just ban it. Anyway...)
Start interval training.
Walk for a minute, jog for a minute, run for a minute, walk for a minute, jog for a minute, etc for 20-30 minutes.
Do 3 times a week. Do on grass or a soft surface with a good pair of shoes.
Long distances walking and jogging.
Do this for an hour to 90 minutes three times a week. Just jog lightly until you can't anymore, then walk until you rested, then jog some more.
Do a weight training routine 2-3 times a week.
It doesn't really matter what you do in this aspect, a standard routine is fine.
You're not trying to get stronger, or put on muscle, you're really just using it to burn calories.
Drink a **** load of water while doing this program. Say 2 litres per day.
Assuming you've got no heart disease or anything nasty (fyi - always check with a doctor before starting...) this is a safe program that should get you close to your goals.
I believe you CAN lose that much weight safely in the amount of time you listed but only if you start now and stick with it rigidly.

Oops, I mis-read...thought he was losing 40lbs in 40 days....25? Yeah, you can do that without health risks, but like FoM said, it's a rough ride and requires discipline.

Willy -
it is only really possible to increase muscle mass and lose fat at the same time if you are a beginner to weight training and are unfit and overweight.
To increase muscle, you need to have a calorie surplus, ie be eating enough food that your body can take care of all its needs plus have extra calories it can use to build muscle.
To lose fat, you need to have a calorie defecit, ie. have slightly less calories than your body needs for full recovery so it uses bodyfat as energy.
Some people are able to lose a small amount of fat and put on a small amount of muscle, but they would be better served by concentrating on one or the other at a time. The Wastrel - So attractive he HAS to be a woman.

Most people tend to go for muscle first...surplus of calories (protein and carbs/fat) and then the added muscle, plus some cardio workin, can eliminate the fat to reveal the muscle in all it's glory.

That's always confused me. Couldn't you just reduce the fat in your diet while still eating a lot?To lose fat, you need to have a calorie defecit, ie. have slightly less calories than your body needs for full recovery so it uses bodyfat as energy.
