There are several existing threads on subject like this. Try putting "conditioning" and/or "knuckle" into the site's search function.
My advice is that it's not worth doing anything active to "condition" your knuckles. Just hit bags and people and do it regularly. If you knuckles hurt, don't hit so hard. -

Pun's right. Doing knuckle push-ups will strengthen your wrists and your fist (maybe), but unless you're young and your bones are soft and malleable, it won't flatten them. It could even cause joint problems.
Just hit stuff, and be sure to treat your hands conditioning/soothing ointments. Nothing mystical, just stuff to help heal.Regards,

Everyone knows that being a martial artist means pain, sacrifice and more excruciating pain.
your knuckles will naturally toughen up and callous over if you are stimulating them on a regular basis, if they don't, just smash them into something hard, like a brick wall.
sure, it will hurt for a while and someone else may have to feed you, but when they heal they will become huge, unfeeling, calcified clubs of doom. -

Make sure you wrap your hands or use gloves, I don't feel anything anymore after working on the bag, but if I wrap my hands poorly sometimes the top layer of skin will peel off. Then it's a bitch to heal as the scab constantly breaks when I make a fist.
Anyway, your knuckles really shouldn't hurt if you punch correctly. Make sure you make impact with the first two knuckles on each hand.
